Objectives: To evaluate the effects of a cell-free supernatant from Lactococcus lactis (CFSM) on performance and diarrhearelated parameters and the presence of F4+ enterotoxigenic E. coli (ETEC) in piglets during post-weaning, and to evaluate the in vitro effect of the CFSM on faeG gene expression in an E. coli F4+. Animals and procedure: In 3 trials with 90 piglets per trial, pigs were assigned to receive a placebo or 1 of 2 CFSM treatments and observed for diarrhea and performance. Fecal swabs were taken to determine the presence of ETEC. Quantitative RT-PCR was used to assess faeG gene expression in E. coli 21259 after treatment with CFSM at 50 mg/mL. Results: The CFSM administered for 14 d at a dose of 24 mg/kg BW (2X) reduced diarrhea-related parameters compared to the placebo. Quantitative RT-PCR showed that, in E. coli 21259 treated with CFSM at 50 mg/mL, expression of the faeG gene was significantly repressed (P < 0.0001) relative to that in the untreated control. Conclusion: The evaluated CFSM reduced the frequency and prevalence of diarrhea in a field situation. The in vitro treatment had an inhibitory effect on the expression of the faeG gene in F4+ E. coli 21259.


Effet d'un surnageant de culture de Lactococcus lactis sur la diarrhée et les paramètres de performance des porcelets en période post-sevrage et sur l'expression du gène faeG in vitro. Objectifs: Évaluer les effets d'un surnageant acellulaire de Lactococcus lactis (CFSM) sur les paramètres de performance et de diarrhée et la présence d'E. coli entérotoxinogène F4+ (ETEC) chez les porcelets en post-sevrage, et évaluer l'effet in vitro du CFSM sur l'expression du gène faeG dans un E. coli F4+. Animaux et procédure: Dans 3 essais portant sur 90 porcelets par essai, les porcs ont reçu un placebo ou 1 des 2 traitements CFSM et ont été observés pour détecter la diarrhée et leurs performances. Des prélèvements fécaux ont été effectués pour déterminer la présence d'ETEC. La RT-PCR quantitative a été utilisée pour évaluer l'expression du gène faeG dans E. coli 21259 après traitement avec CFSM à 50 mg/mL. Objective: The objective of this study was to monitor Streptococcus suis fecal shedding in nursery pigs on a farm with a history of S. suis disease involving serotypes 2 and 9. Animal and procedure: Four cohorts of pigs (n = 480) were monitored from weaning to end of nursery. Rectal swabs from 297 pigs were tested and S. suis serotypes 15 (n = 7), 31 (n = 3), or untypeable (n = 6) isolates were recovered from 16 (5.4%) pigs. Results: There was no significant association between S. suis fecal shedding and diarrhea. Streptococcus suis isolates recovered from pigs euthanized due to neurological signs or severe lameness were serotypes 9 (meninges) and 31 (tonsil) or untypeable (meninges, tonsil). Serotypes 9 (meninges, tonsil), 15 (spleen, tonsil), 16 (tonsil), 29 and 33 (nasal swabs), and untypeable (meninges, tonsil, and lung) isolates were identified in lame pigs. Conclusion and clinical relevance: These results suggest that feces may not be a source of infection for the S. suis serotypes producing disease in pigs; however, the association between S. suis fecal shedding and diarrhea needs further investigation. The coincidence of untypeable isolates in feces from healthy pigs and their isolation from meninges of pigs with neurological signs warrants further investigation to determine the molecular characteristics of those isolates.


Une enquête sur l'excrétion fécale des sérotypes de Streptococcus suis chez les porcelets en pouponnière. Objectif: L'objectif de cette étude était de surveiller l'excrétion fécale de Streptococcus suis chez des porcelets en pouponnière dans une ferme ayant des antécédents de maladie à S. suis impliquant les sérotypes 2 et 9. Animal et procédure: Quatre cohortes de porcs (n = 480) ont été suivies du sevrage jusqu'à la fin de la période en pouponnière. Des écouvillons rectaux provenant de 297 porcs ont été testés et des isolats de S. suis des sérotypes 15 (n = 7), 31 (n = 3) ou non-typables (n = 6) ont été récupérés chez 16 (5,4 %) porcs. Conclusion et pertinence clinique: Ces résultats suggèrent que les matières fécales pourraient ne pas être une source d'infection pour les sérotypes de S. suis produisant des maladies chez les porcs; cependant, l'association entre l'excrétion fécale de S. suis et la diarrhée nécessite des recherches plus approfondies. Animal and objective: The objectives of this study were to produce hematological reference intervals for late-pregnancy sows and to assess whether there were associations between sow hematological end points and the number of stillborn pigs. Procedure: Whole blood was collected from 272 healthy pregnant sows ~1 wk before farrowing, and complete blood (cell) counts and hemoglobin determinations were obtained. Sows were monitored during farrowing, and litter characteristics, including number of stillborn piglets, were recorded. Results and conclusion: Values for red blood cells, hemoglobin, mean corpuscular volume, mean corpuscular hemoglobin, mean corpuscular concentration, platelets, and white blood cells were significantly higher in Parity 1 sows compared to older sows. The mean (± SD) litter size was 14.5 ± 3.4 pigs, with 1.5 ± 2.0 stillborn pigs per litter. There was no significant association between any hematological end point (except white blood cell counts) and the likelihood of a stillborn piglet in a litter. There was a relationship between parity and stillbirth (P < 0.05), with higher-parity sows being more likely than younger sows to produce a stillborn piglet. Although it had been reported that anemic sows were more likely to have stillbirths, perhaps the relatively high mean hemoglobin concentration of sows in this study resulted in the lack of a relationship.


Paramètres hématologiques des truies en fin de gestation et enquête visant à déterminer si ces paramètres sont des prédicteurs de mortinatalités dans un troupeau de truies canadien. Animal et objectif: Les objectifs de cette étude étaient de produire des intervalles de référence hématologiques pour les truies en fin de gestation et d'évaluer s'il existait des associations entre les paramètres hématologiques des truies et le nombre de porcelets mort-nés. Procédure: Du sang total a été collecté auprès de 272 truies gravides en bonne santé environ 1 semaine avant la mise bas, et une numération cellulaire complète et des déterminations d'hémoglobine ont été obtenues. Les truies ont été surveillées pendant la mise bas et les caractéristiques de la portée, y compris le nombre de porcelets mort-nés, ont été enregistrées. Résultats et conclusion: Les valeurs des globules rouges, de l'hémoglobine, du volume corpusculaire moyen, de l'hémoglobine corpusculaire moyenne, de la concentration corpusculaire moyenne, des plaquettes et des globules blancs étaient significativement plus élevées chez les truies de parité 1 que chez les truies plus âgées. Infectious disease events can cause disruptions in service-based and agricultural industries. The list of possible events is long and varies from the incursion or emergence of a reportable animal pathogen to the recently documented interruptions caused by the COVID-19 pandemic. There is a need to develop models that can determine the impact of pathogens and mitigation measures on populations that are not directly affected by the pathogen in the case of a reportable disease, particularly when the health and welfare of these populations could be affected due to resulting disruptions in trade and supply chains. The primary objective of this study was to develop a discrete-event simulation (DES) model of swine production, including pork processing, for scenarios without major disruptions, which could be scaled from the level of an individual farm to the entire province of Ontario, Canada. The secondary objective was to validate the developed simulation against observed farm- and province-level statistics. A weekly discrete-event simulation consisting of 3 connected areas (a sow farm, a pig farm, and abattoirs) was developed using AnyLogic modelling software. Using Mann-Whitney tests, model outputs representative of the standard industry statistics were compared to data from 6 individual farms separately, as well as to provincial data from Ontario. A scalable discrete-event simulation of the swine production system for typical scenarios was accomplished. The model outputs were consistent with individual farm and industry statistics. As such, the model can be used to simulate swine production at distinct levels and could be further modified to represent swine marketing in other provinces or internationally.


Les maladies infectieuses peuvent provoquer des perturbations dans les industries de services et agricoles. La liste des événements possibles est longue et varie de l'arrivée ou de l'émergence d'un agent pathogène animal à déclaration obligatoire aux interruptions récemment documentées causées par la pandémie de COVID-19. Il est nécessaire d'élaborer des modèles permettant de déterminer l'impact des agents pathogènes et des mesures d'atténuation sur les populations qui ne sont pas directement affectées par l'agent pathogène dans le cas d'une maladie à déclaration obligatoire, en particulier lorsque la santé et le bien-être de ces populations pourraient être affectés en raison des conséquences dues aux perturbations du commerce et des chaînes d'approvisionnement. L'objectif principal de cette étude était de développer un modèle de simulation à événements discrets (DES) de la production porcine, y compris la transformation du porc, pour des scénarios sans perturbations majeures, qui pourraient être étendus du niveau d'une ferme individuelle à l'ensemble de la province de l'Ontario, Canada. L'objectif secondaire était de valider la simulation développée par rapport aux statistiques observées au niveau de la ferme et de la province. Une simulation à événements discrets hebdomadaire composée de 3 zones connectées (un élevage de truies, un élevage de porcs et des abattoirs) a été développée à l'aide du logiciel de modélisation AnyLogic. À l'aide des tests de Mann-Whitney, les résultats du modèle représentatifs des statistiques standards de l'industrie ont été comparés aux données de 6 fermes individuelles séparément, ainsi qu'aux données provinciales de l'Ontario. Une simulation à événements discrets évolutive du système de production porcine pour des scénarios typiques a été réalisée. Les résultats du modèle étaient cohérents avec les statistiques individuelles des exploitations et des industries. Ainsi, le modèle peut être utilisé pour simuler la production porcine à des niveaux distincts et pourrait être modifié davantage pour représenter la commercialisation du porc dans d'autres provinces ou à l'échelle internationale.(Traduit par Docteur Serge Messier).

It is unclear if piglets benefit from vaccination of sows against influenza. For the first time, methods of evidence-based medicine were applied to answer the question: "Does vaccine-induced maternally-derived immunity (MDI) protect swine offspring against influenza A viruses?". Challenge trials were reviewed that were published from 1990 to April 2021 and measured at least one of six outcomes in MDI-positive versus MDI-negative offspring (hemagglutination inhibition (HI) titers, virus titers, time to begin and time to stop shedding, risk of infection, average daily gain (ADG), and coughing) (n = 15). Screening and extraction of study characteristics was conducted in duplicate by two reviewers, with data extraction and assessment for risk of bias performed by one. Homology was defined by the antigenic match of vaccine and challenge virus hemagglutinin epitopes. Results: Homologous, but not heterologous MDI, reduced virus titers in piglets. There was no difference, calculated as relative risks (RR), in infection incidence risk over the entire study period; however, infection hazard (instantaneous risk) was decreased in pigs with MDI (log HR = -0.64, 95% CI: -1.13, -0.15). Overall, pigs with MDI took about a ½ day longer to begin shedding virus post-challenge (MD = 0.51, 95% CI: 0.03, 0.99) but the hazard of infected pigs ceasing to shed was not different (log HR = 0.32, 95% CI: -0.29, 0.93). HI titers were synthesized qualitatively and although data on ADG and coughing was extracted, details were insufficient for conducting meta-analyses. Conclusion: Homology of vaccine strains with challenge viruses is an important consideration when assessing vaccine effectiveness. Herd viral dynamics are complex and may include concurrent or sequential exposures in the field. The practical significance of reduced weaned pig virus titers is, therefore, not known and evidence from challenge trials is insufficient to make inferences on the effects of MDI on incidence risk, time to begin or to cease shedding virus, coughing, and ADG. The applicability of evidence from single-strain challenge trials to field practices is limited. Despite the synthesis of six outcomes, challenge trial evidence does not support or refute vaccination of sows against influenza to protect piglets. Additional research is needed; controlled trials with multi-strain concurrent or sequential heterologous challenges have not been conducted, and sequential homologous exposure trials were rare. Consensus is also warranted on (1) the selection of core outcomes, (2) the sizing of trial populations to be reflective of field populations, (3) the reporting of antigenic characterization of vaccines, challenge viruses, and sow exposure history, and (4) on the collection of non-aggregated individual pig data.

Introduction: Group A rotaviruses are major pathogens in causing severe diarrhea in young children and neonates of many different species of animals worldwide and group A rotavirus sequence data are becoming increasingly available over time. Different methods exist that allow for rotavirus genotyping, but machine learning methods have yet to be explored. Usage of machine learning algorithms such as random forest alongside alignment-based methodology may allow for both efficient and accurate classification of circulating rotavirus genotypes through the dual classification system. Methods: Random forest models were trained on positional features obtained from pairwise and multiple sequence alignment and cross-validated using methods of repeated 10-fold cross-validation thrice and leave one- out cross validation. Models were then validated on unseen data from the testing datasets to observe real-world performance. Results: All models were found to perform strongly in classification of VP7 and VP4 genotypes with high overall accuracy and kappa values during model training (0.975-0.992, 0.970-0.989) and during model testing (0.972-0.996, 0.969-0.996), respectively. Models trained on multiple sequence alignment generally had slightly higher overall accuracy and kappa values than models trained on pairwise sequence alignment method. In contrast, pairwise sequence alignment models were found to be generally faster than multiple sequence alignment models in computational speed when models do not need to be retrained. Models that used repeated 10-fold cross-validation thrice were also found to be much faster in model computational speed than models that used leave-one-out cross validation, with no noticeable difference in overall accuracy and kappa values between the cross-validation methods. Discussion: Overall, random forest models showed strong performance in the classification of both group A rotavirus VP7 and VP4 genotypes. Application of these models as classifiers will allow for rapid and accurate classification of the increasing amounts of rotavirus sequence data that are becoming available.

Objectives: Pigs often experience slow growth and enteric disease problems during the early post-weaning period. The objectives were to evaluate effects of a live oral E. coli vaccine on post-weaning diarrhea under on-farm conditions and to assess impacts of diet composition on growth and enteric health in the early nursery phase of production. Procedures and results: In a series of trials, even when enterotoxigenic E. coli (ETEC) was present, post-weaning diarrhea was usually associated with other causes and not ETEC. Therefore, an E. coli vaccination program had no benefit in either reducing clinical signs of diarrhea or in improving growth rate of pigs in the nursery. Conversely, under the same conditions, feeding programs affected both clinical signs of diarrhea and growth rate. Pigs fed a 4-phase program that transitioned from a diet containing a relatively high proportion of animal-based protein to a feed consisting of plant-based protein performed better than pigs receiving lower-complexity diets. However, there was evidence of compensatory growth in pigs fed low-complexity diets, albeit not consistently in all trials. Conclusion and clinical relevance: It was concluded that early nursery diet can help to reduce post-weaning diarrhea and improve growth performance.

Les objectifs étaient d'évaluer les effets d'un vaccin oral vivant contre E. coli sur la diarrhée post-sevrage dans des conditions à la ferme et d'évaluer les impacts de la composition du régime alimentaire sur la croissance et la santé entérique au début de la phase de production en pouponnière. Procédures et résultats: Dans une série d'essais, même en présence d'E. coli entérotoxinogène (ETEC), la diarrhée post-sevrage était généralement associée à d'autres causes et non à l'ETEC. Par conséquent, un programme de vaccination contre E. coli n'a eu aucun effet bénéfique sur la réduction des signes cliniques de diarrhée ou sur l'amélioration du taux de croissance des porcs en pouponnière. Inversement, dans les mêmes conditions, les programmes d'alimentation ont affecté à la fois les signes cliniques de diarrhée et le taux de croissance. Les porcs nourris avec un programme en quatre phases qui sont passés d'un régime contenant une proportion relativement élevée de protéines animales à un aliment composé de protéines végétales ont obtenu de meilleurs résultats que les porcs recevant des régimes moins complexes. Cependant, il y avait des preuves d'une croissance compensatoire chez les porcs nourris avec des régimes à faible complexité, mais pas de manière constante dans tous les essais. In this study, we assessed the efficacy of a novel Bacillus subtilis probiotic in improving growth performance and gut responses in comparison to pharmacological zinc oxide (ZnO) in nursery pigs. A total of 96 piglets were randomly assigned to four groups: Negative control (NC), Positive control (PC, 3000 mg Zn /kg feed), B.subtilis low dose (BS9-L, 2 × 107 CFU/pig) and B.subtilis high dose (BS9-H, 2 × 109 CFU/pig). Growth performance, diarrhea rate, gut mucosal gene expression and fecal microbial populations were evaluated. B.subtilis administration did not improve piglet bodyweight. BS9-L showed (P < 0.05) higher average daily gain (ADG) in Period 2 (D14-D28). BS9 groups had (P < 0.001) lower feed conversion ratio (FCR) in Period 2 (D14-D28) and overall. Like the ZnO-group, BS9 groups had lower (P < 0.01) diarrhea rate. A significant reduction (P < 0.05) in fecal E. coli, total coliforms, and an increase in lactic acid bacteria and Bacillus spp. in BS9 groups was observed. BS9 group had reduced (P < 0.05) mRNA levels of intestinal IL-8 and higher levels of MUC-1 and occludin and TJP-1 compared to negative control. These findings suggest that probiotic BS9, may promote growth performance, and ameliorate various indicators of intestinal health in piglets. Hence, it may serve as a prospective alternative to ZnO growth promoter in commercial swine production.

Eighty-eight gilts [initial body weight (BW) 49.8 ± 0.8 kg] were recruited to determine the effects of moderate energy and protein restriction during the development period on changes in BW and backfat depth (BF) and subsequent lactation performance. Gilts were randomly assigned to one of four feeding programs: 1) standard commercial diet fed ad libitum (CON), 2) standard commercial diet fed 10% or 3) 20% below ad libitum, or 4) a high-fiber diet fed ad libitum [2.5 times more fiber (neutral detergent fiber) than the commercial diet to dilute net energy and crude protein by approximately 20% and 13%, respectively; FIB]. The gilts were housed individually and received the feeding programs between 90 and 190 (breeding) d of age and standard gestation and lactation diets thereafter. Litters were standardized to 12 ± 1 pigs within 48 h of farrowing; weaning occurred at 20.0 ± 0.4 d of age. Gilts that received the 20% restricted program had lower overall average daily feed intake (ADFI) during the development period (2.64 ± 0.04 kg; P < 0.05) versus all other feeding programs and gilts that received the 10% restricted program had lower ADFI than FIB and CON, which were not different (2.96 vs. 3.44 and 3.47 ± 0.04 kg for 10%, FIB and CON, respectively; P < 0.05). Plasma free fatty acid (FFA) concentrations on day 180 of age were lower for gilts that received the 10% and 20% restricted programs compared to gilts that received the FIB and CON programs, which were not different (97 and 86 vs. 220 and 149 ± 29 µEq/L, respectively; P < 0.05). Plasma concentrations of glucose, urea, prolactin, and IGF-1 were not different among feeding programs on day 180 of age. At breeding, gilts that received the FIB and 10% programs had lower BW and BF versus CON (145.7 and 144.8 vs. 155.2 ± 0.9 kg and 14.4 and 14.8 vs. 16.5 ± 0.2 mm for BW and BF, respectively; P < 0.05) but greater BW than gilts that received the 20% restricted program (137.9 kg; P < 0.05). The BW and BF of gilts did not differ at the end of gestation or at weaning. The ADFI of sows during lactation and offspring birth weight and growth rate during lactation and the 5-wk nursery period were not influenced by gilt development feeding program. Therefore, a high-fiber feeding program could be used in group-housing gilt-development scenarios, where feed is offered ad libitum, to control BW and BF prior to breeding without influencing milk production in the subsequent lactation period.


Gilt development feeding programs are important to ensure that the female grows at an appropriate rate, without becoming overconditioned or developing locomotion issues. Moreover, between 90 d of age and puberty is the first phase of rapid mammary development, which is necessary to support milk production after farrowing and which can be negatively impacted by reduced energy and protein intakes. Increasing the dietary fiber content is a means to reduce energy intake, to slow growth rates, and to minimize excessive body fat gains. In this study, it was demonstrated that providing a high-fiber feeding program during the gilt development period controlled body weight (BW) and backfat thickness at breeding, while subsequent lactation performance was not impacted. Therefore, a high-fiber feeding program could be used in group-housing gilt-development scenarios where feed is offered ad libitum, in order to control BW and fatness prior to breeding, without influencing milk production in the subsequent lactation period.

The goal of this study was to determine if seasonality of rotavirus A, B, and C infection is present in Ontario and Quebec swine herds by investigating submissions to a diagnostic laboratory. Samples (N = 1557) within 755 case submissions from Canadian swine herds between 2016 and 2020 were tested for rotaviruses A, B, and C using a real-time polymerase-chain reaction assay and described. Data from Ontario and Quebec were additionally analyzed using boxplots, 6-week rolling averages, time-series decomposition, and negative binomial regression models. Percentage positivity of submissions for rotaviruses A, B, and C were discovered to be highest in nursery/weaner (n = 100, 94.0%, 60.0%, 80.0%) and grower/finisher (n = 13, 84.6%, 46.2%, 61.5%) pigs and lowest in gilt/sow (n = 45, 68.9%, 20.0%, 40.0%) and suckling pigs (n = 102, 67.6%, 10.8%, 38.2%), respectively. The most common combination of rotavirus at the sample level was AC (n = 252, 17%) and ABC (n = 175, 23.2%) at the submission level. Percent positivity for rotavirus A, B, and C across all Canadian provinces included in the study were 69.9%, 32.6%, and 53.1%, respectively. Descriptive analysis suggested little to no evidence of seasonal patterns, although a spike in November was seen in the monthly total submissions and monthly total positive submissions. Statistically, the overall month effect could not be identified as statistically significant (P > 0.05) for any of the evaluated submission counts. Overall, there was no evidence supporting seasonality of rotavirus within Ontario and Quebec swine herds between 2016 and 2020.


Le but de cette étude était de déterminer si la saisonnalité de l'infection à rotavirus A, B et C est présente dans les troupeaux de porcs de l'Ontario et du Québec en examinant les soumissions à un laboratoire de diagnostic. Des échantillons (N = 1557) de 755 cas soumis de troupeaux de porcs canadiens entre 2016 et 2020 ont été testés pour les rotavirus A, B et C à l'aide d'un test de réaction d'amplification en chaîne par polymérase en temps réel et décrits. This case study describes a severe tail-biting event on a multi-site swine operation in Ontario and outlines the management strategies implemented in an attempt to control the problem. An established social order was clearly present before the tail-biting event occurred. Over 40% of tail-docked pigs in 3 of 8 grower-finisher barns were severely affected, leading to higher mortality and increased numbers of pigs re-housed in hospital pens. Environmental factors, management practices, and animal health in the barns experiencing the tail-biting event are described, including detection of the mycotoxin deoxynivalenol in corn at > 2 ppm. Changes implemented in response to tail-biting included altering the phase-feeding schedule, adding enrichment devices, and increasing surveillance. The subsequent cohort of pigs was followed through the finisher barns and did not engage in the same severity or prevalence of tail-biting as the previous cohort of pigs which experienced the tail-biting event. Key clinical message: No single factor was identified as the initiating cause for the severe tail-biting event. The subsequent cohort of pigs in 4 barns of the same operation were monitored for tail-biting from entry until market, and the incidence of tail-biting was very low.

The objective of this systematic review and meta-analysis was to identify the benefits and possible adverse side effects of oxytocin use during farrowing. Randomized controlled trials that were published in English within the last 50 years were eligible for inclusion. Eligible research needed to contain the PICO elements: population (P)-sows at farrowing; intervention (I):-oxytocin given to sows-comparator (C): sows at farrowing not given oxytocin, as well as sows given different dosages and/or different timing of administration; and outcomes (O):-stillbirths, sow mortality, and piglet viability. Four bibliographic databases were used: PubMed, CAB Direct, Web of Science Core Collection, and ProQuest Dissertations, and Theses Global. In addition, we performed a manual search of the table of contents in the American Association of Swine Veterinarians database for relevant conference proceedings and reports. To assess the risk of bias at the study level, a modified version of the Cochrane 2.0 ROB was used. Meta-analyses were performed to examine the average stillbirth rate, farrowing duration, and birth interval between piglets using random-effect standardized mean difference (SMD) models. To explore heterogeneity, a sub-group analysis was performed on the objectives of the study, dose, time, and route of administration. Of the 46 studies eligible for meta-analyses, only 25 had sufficient information. The pooled analyses of the random effect model demonstrated that the average number of stillborn pigs was lower in the comparator group (SMD = 0.23; CI95% = 0.1, 0.36), and both the farrowing duration (SMD = -8.4; CI95% = -1.1, -0.60) and the birth interval between piglets (SMD = -1.41; CI95% = -1.86, -0.97) were shorter in the oxytocin group. The majority of the studies had an overall risk of bias of 'some concerns'. It was concluded that the use of oxytocin increases the overall number of stillborn piglets, but decreases the farrowing duration and time interval between piglets. However, future studies should focus on the effect of oxytocin on the experience of dystocia among sows.

Objective: The objective was to evaluate the pharmacokinetics of compounding non-steroidal anti-inflammatory drugs (NSAIDs) meloxicam or flunixin meglumine with iron dextran (ID) in piglets. Animal: Forty piglets (8 d of age) were randomly allocated into 5 groups (8 piglets/group) and received 1 intramuscular injection in the neck of the following treatments: flunixin meglumine (2.2 mg/kg) administered alone (F) or mixed with ID (F+ID); or meloxicam (0.4 mg/kg) administered alone (M) or mixed with ID (M+ID); or ID alone. Procedure: Blood samples were collected via indwelling jugular catheters at pre-dose, and 10, 20, 30, 45, and 60 min, and 2, 4, 8, 12, 24, 36, 48, and 72 h post-treatment to determine plasma NSAIDs concentrations using liquid chromatography-tandem mass spectrometry. Pharmacokinetic parameters for plasma meloxicam and flunixin meglumine concentration-time profiles were determined for each piglet using noncompartmental analysis approaches. Statistical analyses were performed using SAS software with significance set at P < 0.05. Results: The AUC0-tlast, AUC0-∞, Cmax, and relative bioavailability values in the M+ID and F+ID groups were lower than corresponding M and F groups. The M+ID group elimination half-life was lower, whereas λz and tmax values were greater than the corresponding M group. Conclusion: Relative bioavailability of meloxicam and flunixin meglumine were reduced when compounded with ID in the same bottle and administered to piglets. Clinical relevance: Further research is warranted to evaluate if decreased NSAID exposure when compounded with ID alters analgesic efficacy or drug residue depletion.

Body lesions, resulting from tail-biting and ear-biting, can result in decreased health and welfare in pigs. Tryptophan, an indispensable amino acid, is needed to support protein deposition, and the synthesis of the neurotransmitter serotonin, which is important to mood, sleep-wake and eating patterns and might play a role in aggression and abnormal behavior. Two randomized block design studies were conducted to assess the influence of varying dietary tryptophan levels on aggression and abnormal behavior in 8-week-old pigs. Six diets were formulated which met or exceeded all nutrient requirements yet differed according to the dietary tryptophan content. The first study included control (100% standardized ileal digestible tryptophan), supplemented (175% standardized ileal digestible tryptophan), and supplement-plus (250% standardized ileal digestible tryptophan) experimental diets, while the second study included deficient (80% standardized ileal digestible tryptophan), adequate control (105% standardized ileal digestible tryptophan), and extra-tryptophan (130% standardized ileal digestible tryptophan) experimental diets. Concentrations of plasma tryptophan and large neutral amino acids (tyrosine, isoleucine, leucine, valine, and phenylalanine) were analyzed using ultra-performance liquid chromatography and the tryptophan to large neutral amino acid ratio was calculated. Analysis for time active, lying, and engaging in aggressive interactions was carried out using 10-min scan samples to determine behavioral time budgets of the pigs on different experimental diets. Pigs fed diets with supplemented tryptophan had higher concentrations of both plasma tryptophan and tryptophan to large neutral amino acid ratio compared to the pigs fed the control diet (P < 0.05) in the first study, while no significant differences were detected for plasma tryptophan or the tryptophan to large neutral amino acid ratio in the second study. Diet did not have an effect (P > 0.05) on weight, feed intake or behavior throughout the studies. The results suggest that an increase in dietary tryptophan relative to large neutral amino acids, fed for 29 days, impacts circulating plasma tryptophan and therefore, serotonin concentrations in the pig. Despite an increase in circulating plasma tryptophan in response to an increase in dietary tryptophan in the first study, we failed to see an impact of the dietary treatment on body, tail and ear-biting behavior under the conditions studied.

In Canada, piglets receive analgesia to control pain after surgical castration. There is interest in examining the potential to mix non-steroidal anti-inflammatory drugs with iron dextran prior to injection to minimize piglet handling and labor. The objective of this study was to compare pharmacokinetics and the relative bioavailability of ketoprofen given alone (3.0 mg/kg IM) versus the same dose of ketoprofen mixed with iron dextran (52.8 mg/kg IM) (ketoprofen + iron dextran) before injection in piglets. Piglets 8 to 11 d old were allocated into 2 treatment groups (n = 8/group). Plasma drug concentrations were measured using mass spectrometry at 13 time points after injection. No significant differences were detected between the 2 groups when examining pharmacokinetic parameters (e.g., Cmax, Tmax, AUC) or relative bioavailability for either S- or R-ketoprofen enantiomers (P > 0.05). However, pain control efficacy and food safety studies of these formulations are required to further examine this practice.

Tail-biting is globally recognized as a welfare concern for commercial swine production. Substantial research has been undertaken to identify risk factors and intervention methods to decrease and understand this vice. Tail-biting appears to be multifactorial and has proven difficult to predict and control. The primary objective of the scoping review was to identify and chart all available literature on the risk factors and interventions associated with tail-biting in pigs. A secondary objective was to identify gaps in the literature and identify the relevance for a systematic review. An online literature search of four databases, encompassing English, peer-reviewed and grey literature published from 1 January 1970 to 31 May 2019, was conducted. Relevance screening and charting of included articles were performed by two independent reviewers. A total of 465 citations were returned from the search strategy. Full-text screening was conducted on 118 articles, with 18 being excluded in the final stage. Interventions, possible risk factors, as well as successful and unsuccessful outcomes were important components of the scoping review. The risk factors and interventions pertaining to tail-biting were inconsistent, demonstrating the difficulty of inducing tail-biting in an experimental environment and the need for standardizing terms related to the behavior.

Porcine reproductive and respiratory syndrome virus (PRRSV) is one of the most important swine viruses globally, including in Ontario, Canada. Understanding the evolution and relation of the various PRRSV genotypes in Ontario can provide insight into the epidemiology of the virus. The objectives of this study were to i) describe the variability of PRRSV genotypes in Ontario swine herds, and ii) evaluate possible groupings based on PRRSV genomic data. Virus open reading frame 5 (ORF-5) sequences collected from 2010 to 2018 were obtained from the Animal Health Laboratory, University of Guelph and Bayesian phylogenetic models were created from these. The PRRSV population of Ontario was then categorized into 10 distinct clades. Model comparisons indicated that the model with a constant population assumption fit the data best, which suggests that the net change in the PRRS virus variation of the entire population over the last decade was low. Nonetheless, viruses grouped into individual clades showed temporal clustering during distinct time intervals of the entire study period (P < 0.01).

While heart failure is a primary cause of death for many in-transit-loss (ITL) pigs, the underlying cause of these deaths is not known. Cardiomyopathies are considered a common cause of heart failure in humans and often have a genetic component. The objective of this study was to determine if genes associated with cardiomyopathies could be identified in ITL pigs. Samples from the hearts of pigs that died during transport to an abattoir in Ontario, Canada were collected and genotyped along with samples from pigs that did not die during transport (ILT hearts: n = 149; non-ITL/control hearts: n = 387). Genome-wide analyses were carried out on each of the determined phenotypes (gross cardiac lesions) using a medium density single nucleotide polymorphism (SNP) chip and 500 kb windows/regions for analysis, with 250 kb regions of overlap. The distribution derived by a multidimensional scaling (MDS) analysis of all phenotypes demonstrated a lack of complete separation between phenotypes of affected and unaffected animals, which made diagnosis difficult. Although genetic differences were small, a few genes associated with dilated cardiomyopathy (DCM) and arrhythmogenic right ventricular cardiomyopathy (ARVM) were identified. In addition, multiple genes associated with cardiac arrhythmias and ventricular hypertrophy were identified that can possibly result in heart failure. The results of this preliminary study did not provide convincing evidence that a single, heritable cardiomyopathy is the cause of heart failure in ITL pigs.

L'objectif de cette étude était de déterminer si des gènes associés aux cardiomyopathies pouvaient être identifiés chez les porcs ITL. Des échantillons de coeurs de porcs morts pendant le transport vers un abattoir en Ontario, au Canada, ont été prélevés et génotypés avec des échantillons de porcs qui ne sont pas morts pendant le transport (coeurs ILT: n = 149; coeurs non ITL/témoins: n = 387). Des analyses à l'échelle du génome ont été effectuées sur chacun des phénotypes déterminés (lésions cardiaques macroscopiques) en utilisant une puce de polymorphisme nucléotidique unique (SNP) de densité moyenne et des fenêtres/régions de 500 kb pour l'analyse, avec des régions de chevauchement de 250 kb. La distribution dérivée par une analyse de mise à l'échelle multidimensionnelle (MDS) de tous les phénotypes a démontré un manque de séparation complète entre les phénotypes des animaux affectés et non affectés, ce qui a rendu le diagnostic difficile. Bien que les différences génétiques soient minimes, quelques gènes associés à la cardiomyopathie dilatée (DCM) et à la cardiomyopathie arythmogène ventriculaire droite (ARVM) ont été identifiés. De plus, plusieurs gènes associés aux arythmies cardiaques et à l'hypertrophie ventriculaire ont été identifiés, pouvant éventuellement entraîner une insuffisance cardiaque. Les résultats de cette étude préliminaire n'ont pas fourni de preuves convaincantes qu'une seule cardiomyopathie héréditaire est la cause de l'insuffisance cardiaque chez les porcs ITL.(Traduit par Docteur Serge Messier).

Influenza A viruses (IAVs) are common causes of respiratory infection in pigs. The objective of this study was to characterize the circulation of IAVs between weaning and market age on the basis of development of antibody response and molecular epidemiology of detected viruses. Two batches of weaned pigs were followed in the nursery and finisher barns with a sample of 81 and 75 pigs. Nasal swabs and blood samples were collected from individual pigs for virological and serological analyses. A H3N2 subtype virus, of cluster IV, was detected in Study 1, with a maximum of 97.9% identity to HA gene of viruses previously isolated in Ontario. In Study 2, a H1N1 subtype virus, of 2009 H1N1 pandemic lineage, was detected, with a maximum of 97.8% identity to HA gene of viruses previously isolated in Ontario. On the basis of HA gene, it was observed that pigs were being detected with the same virus over time. The existence of antibody titers for IAV other than the isolated one confirmed that more than one subtype can circulate in the same population. In Study 1, pigs with higher numbers of IAV detection had lower serological titers for the same virus that was confirmed to circulate in the nursery (P < 0.01). Thorough knowledge of all endemic viral strains is fundamental for development of infection and disease control, particularly in complex production systems. This may include consideration of sampling and testing strategies which could detect circulation of all IAV variants, even if they have low prevalence.

Streptococcus suis naturally colonizes the upper respiratory tract of pigs and can lead to severe disease conditions. Although there are several serotypes associated with disease, untypable isolates have also been observed. The objective of this study was to investigate the relatedness of untypable S. suis isolates detected in clinical cases and healthy pigs in Ontario, Canada, and their relation to typing serotypes. One hundred fifty-six isolates obtained from 33 cases and 26 farm-and-pen-matched control pigs were sequenced using Illumina HiSeq sequencing. Protein sequences of the capsular polysaccharide genes (cps) were identified and analyzed using a maximum likelihood tree. Among the 27 untypable isolates, 3 were from systemic sites of cases and 13 and 11 were from upper respiratory sites of cases and controls, respectively. One hundred fifty-six isolates were grouped into 17 distinct groups based on the cps gene tree. Isolates from these 17 distinct individual cps groups were distributed among a minimum of one farm and maximum of eight farms. Untypable isolates were detected in 12 of those groups and each cps group had untypable isolates present amongst multiple farms. Interestingly, the three systemic untypable isolates not only coexisted with other serotypes found in the same location of the same pigs but were also found among different cps groups. These isolates are of interest and warrant further investigation. Overall, a wide diversity of S. suis among untypable isolates was observed in this study.
